Sapreme’s mission is to develop next-generation macromolecule therapeutics by circumventing endosomal entrapment, thereby enhancing target engagement. The company’s proprietary endosomal escape platform improves the therapeutic window by enabling access to intracellular targets with minimal toxicity. This approach is applied for Sapreme’s internal pipeline and is available for partnering, without limitation to biologic modality or indication. Our solution to endosomal entrapment has the potential to re-open myriad drug development pathways for macromolecule therapeutics lacking efficacy and to explore new targets considered “undruggable” by permeabilizing endo-lysosomal membranes, allowing trapped macromolecules to be released, increasing target engagement without negatively impacting the plasma membrane integrity.

In the course of 2022, InteRNA concluded the first part of its Phase 1a clinical trial with lead candidate INT-1B3 showing a positive safety profile. Unfortunately, due to lack of capital InteRNA was not able to start the second part of this trial for which the necessary regulatory approvals were in place. Despite also the already obtained clearance from the FDA for the Phase 1b/2a trial, the outlook on a combinatory trial with one of the leading PD1 inhibitors in the immune-oncology marketplace and a positive FTO analysis, InteRNA has not been able to attract further funding in the distressed financing climate and was forced to cease operations at the end of Q1-2023. Here we wish to extend our sincere gratitude to the participating patients and their families, as well as our employees, board members, clinical and scientific advisors, investors and partners for their support throughout our journey. It is a pity that INT-1B3 with its promising and unique Mode of Action did not get the chance to be validated in a clinical trial for its efficacy.
