Zag Bio is a group of bold scientists and expert drug developers, formed to pioneer thymus-targeted medicines to treat and prevent autoimmune diseases. We have discovered a novel therapeutic approach to restore central immune tolerance by creating bifunctional antibodies that deliver self-antigens to specialized antigen-presenting cells in the thymus. This enables us to harness the body’s natural process for training immune cells to recognize and tolerate self, halting or preventing autoimmune attacks on the body’s own tissues. Zag Bio’s pipeline includes its lead program ZAG-101, a bifunctional antibody that delivers pancreatic beta cell antigens to the thymus to treat Type 1 diabetes, as well as discovery programs for thymus-targeted therapies to address other autoimmune diseases.
Zag Bio is located in Cambridge, MA, and is supported by a broad syndicate of life sciences investors. Zag Bio’s team is built on the mission of improving the lives of patients by charting a new direction for better therapies for autoimmune diseases, based on unique insights and understanding of the biology of the thymus.

Pandion Therapeutics is a biotechnology company developing bispecific antibody therapeutics to achieve localized immunomodulation at the site of disease for durable, tissue-specific treatment of patients with autoimmune and inflammatory disease and organ transplants. Our localized approach represents a significant change from the systemic immunosuppression of conventional medicines, which often have dose-limiting safety concerns and can cause serious infections and cancer. Furthermore, through tissue-specific therapeutic targeting, Pandion’s approach has the potential to more effectively induce and sustain response and remission in patients with many autoimmune and inflammatory conditions. We aim to shift the paradigm and create a new generation of drugs with transformative efficacy and improved safety.

Totient is an AI-driven biotechnology company leveraging tertiary lymphoid structures (TLSs) to identify novel tissue-specific antigens and develop matching high-affinity antibody therapeutics. Totient reconstructs antibodies from tissues affected by autoimmunity, infections, and cancer collected from patients experiencing exceptional immune responses. Beginning with a population of over 50,000 samples, we have reconstructed antibodies against known and novel extracellular oncology targets: GAGE1, GAGE2A, ANXA1, C4BPB, IL14A, BIRC7, HCLS1, GPR83, and others undisclosed.
