Inexia is developing intranasal orexin receptor agonists and positive modulators for the treatment of narcolepsy, a rare neurological condition that affects the brain's ability to regulate the normal sleep-wake cycle Orexin, also called ‘hypocretin’, is a key regulator of wakefulness and REM sleep. We aspire to develop innovative medicines that address the full spectrum of orexin dysfunction in disease areas where there is high unmet clinical need. Inexia is designing novel intranasal small molecule and peptide-based agonists and positive modulators, which would influence orexin neurotransmission differently. Orexin agonists and positive modulators have the potential to treat a wide range of disorders which are inadequately treated today, most notably Narcolepsy Type 1 (NT1) which is caused by profound loss of orexin-producing neurons. NT1 is a chronic rare disease that usually starts during adolescence and is estimated to effect over 3 million people worldwide. Orexin agonists and positive modulators will directly address the underlying disease pathology of NT1, and may have the potential for greater efficacy and tolerability than current therapies. Inexia is founded by Medicxi in collaboration with Sosei Heptares and leverages their unique GPCR structure-based drug design (SBDD) capability for the discovery and development of promising drug candidates. Inexia collaborates closely with Orexia, founded by Medicxi in collaboration with Sosei Heptares for developing oral orexin receptor agonists and positive modulators.
