Graffinity SCREENING SERVICES find small molecules that bind to proteins; a staring point for DISCOVERY of drugs or affinity ligands for protein purification. The staff are surface plasmon resonance experts advising on implementation of SPR.
Higher throughput screens employ Graffinity microarrays - produced in-house – containing 9,216 immobilized small molecule ligands - with instruments not available elsewhere. Medium throughput tasks such as fragment library screens use microfluidic SPR Biacore® systems.
DRUG DISCOVERY – screening platform for fragment & lead-like hits
Small molecule drug discovery is served by Graffinity & its parent NovAliX through a unique integration of screening, medicinal chemistry & structural biology. The Graffinity library of 116,000 compounds has provided a starting point for chemical synthesis for over 90% of > 110 projects. A powerful reservoir of data concerning the binding properties of novel compounds has been created.
The high sensitivity is particularly well suited for fragment based drug discovery, FBDD. Fragments (MW < 300) represent > 20% of the library. Recently great emphasis has been placed upon improving the quality of the library though the addition of compounds with favorable medicinal chemistry properties. The technology is equally applicable to target based discovery for agrochemicals, food flavors, fragrances & cosmetics.
AFFINITY PURIFICATION: PROTEINS/biologics/enzymes - downstream processing
A screening service discovers novel ligands for novel proteins. Development projects for antibody purification include: 1) a generic Fab binder, 2) HCP capture for reverse purification or polishing 3) an Fc binder with improved properties versus protein A. The probability of finding useful affinity is high and any purification systems will be novel, patentable. http://tiny.cc/SlideShareAffLig
